Cytotoxic effects of organochlorine insecticides in Baby Hamster Kidney (BHK 21 C13) cell line
The acute toxicity evaluation and safety assessment of new chemicals and substances like pesticides and the extrapolation of those data into human risk assessment, after entering via food or water chain, requires a lot of expensive and time-costing tests in animal species. In order to reduce the number of experimental animals in acute toxicity testing assays on cell culture systems are developing. In the present work BHK 21 C13 cell line was used as a test-system for the determination of organochlorine insecticides heptachlor, lindane and aldrin toxicity measured by Trypan blue exclusion method. Results showed, that compared to control values, heptachlor in concentrations 5-125 microg/ml induced cytotoxicity effects in the the range 15-92% ; lindane in concentrations 10-100 microg/ml for 23-78% and aldrin in concentrations 3-125 microg/ml for 21-100%, respectively. IC_50 values for heptachlor, lindane and aldrin were derived graphically and were 34.4 ; 35.8 and 11.0 microg/ml, respectively. Cytotoxicity data for heptachlor, lindane and aldrin were used in prediction of in vivo acute toxicity by applying the linear regression between IC_50 and oral LD_50 values of the Register of Cytotoxicity.
