Beta-glucan from brewers yeast and algae Euglena gracilis

Recently beta-glucan attracts considerable research interest mostly because of its positive effect on human health. beta-glucan has capability to inhibit the growth of bacteria, molds and various tumors, to stimulate immune system, and to lower cholesterol level in blood. Sulfated derivatives of beta-glucan significantly inhibit the cytophatic effect of human immunodeficiency virus (HIV-1, HIV-2) and the expression of HIV antigen in human peripheral blood mononuclear cells. Main sources of beta-glucan in nature are crops (barley, rice, oat and wheat) and microorganisms (yeast and algae). Relatively low beta-glucan content in crops is the main reason that microbial biomass is the most interesting source of beta-glucan for large scale production. Brewer^, s yeast, as a by-product of beer production, contains beta-glucan in cell wall and consequently relatively complex extraction procedure is required for successful beta-glucan isolation. Furthermore, beta-glucan content in yeast cell wall is variable and it is mainly effected by yeast growth and fermentation conditions. In opposite to the yeast, unicellular algae Euglena gracilis has capability to accumulate intracellularly large amounts of 1, 3-beta-glucan (paramylon) when it grows in heterotrophic conditions. Therefore, recovery of beta-glucan from E. gracilis is not so complex process compared to the beta-glucan isolation from yeast. This work deals with: 1. Research on the most adequate procedure for beta-glucan isolation from brewer^, s yeast, 2. Cultivation of Euglena gracilis on different complex media and 3. Isolation of beta-glucan from E. gracilis.
