The role of SWI/SNF and histone acetiltransferase activity of SAGA complex in transcriptional regulation at the yeast PHO84 promoter

The yeast PHO5 and the PHO8 genes have become a well known system to investigate the role of chromatin structure in transcriptional regulation. Both genes are regulated in response to the phosphate concentration by the same specific transcription factor, Pho4. Upon induction by phosphate starvation both promoters undergo characteristic chromatin remodeling. Chromatin remodeling at the PHO8 promoter essentially requires the SWI/SNF and SAGA complexes at a step subsequent to the binding of the transcription factor Pho4. The rate of chromatin remodeling step at the PHO5 promoter is strongly delayed in the absence of SWI/SNF or SAGA complex, but in contrast to PHO8 promoter they are not essentially required for remodeling. Among the PHO genes the PHO84 gene, encoding a Pi transporter, is the strongest promoter among the PHO genes, containing five Pho4 binding sites. In this work we investigate chromatin structure transition upon induction of the POH84 promoter and requirement of chromatin remodeling machines SWI/SNF and SAGA complexes in activation of different PHO84 promoter derivatives containing mutated Pho4 binding sites.
