Influence of atrazine on 5-alpha-DHT regulated mechanisms in rat ventral prostate

Environmental pollutants, like pesticides could interfere with physiological processes in mammalian reproductive system. Effect of atrazine, s-triazine herbicide, on 5-alpha-DHT-receptor complex formation in prostate nuclei was examined. 5-alpha-DHT-receptor complex in rat prostate nuclei was characterized by 12,6 % SDS-PAGE. Specific binding of 3H-DHT on protein molecule in rat prostate nuclei was determined at 20 kD, 43 kD and 67 kD. The largest inhibition after in vitro addition or after i.p. treatment with atrazine was found at Mr 43 kD. The binding of 3H-DHT to nuclear receptor was decerased for ~79% at Mr 43 kD under atrazine concentration of 0.7 mmol. After in vivo atrazine treatment ( 60 mg/kg b.w., i.p., daily, over 10 days) 3H-DHT-receptor complex formation at Mr 43 kD was inhibited for 42% and at the higher dose (120 mg/kg b.w., i.p. daily, over 10 days) the inhibition was 63%. By introducing of saturation analysis the number of specific binding sites on the receptors in prostate nuclei and the dissociation constant (Kd) of 5-alpha-DHT-receptor complex were determined. After i.p. treatment with 120 mg atrazine/kg b.w. daily over 10 days or in vitro addition of 0.7 mmol atrazine Kd-values were at the same range (10^-12 M), but the number of specific binding sites for 5-alpha-DHT on receptor molecule decreased for ~50 %. The results show that atrazine has a crucial inhibitory effect for 5-alpha-DHT-receptor complex formation in rat prostate nuclei.
