Different patterns of killing and mutagenesis in Salmonella typhimurium induced by hydrogen peroxide

H2O2 is one of the active oxygen species which are correlated with several degenerative diseases in mammals such as cancer, aging and arhritis. In bacteria, oxidative stress mediated by exogenous H2O2 can be lethal or mutagenic. Toxic and mutagenic properties of H2O2 most probably are derived through the action of hydroxyl radical (OH-) which is highly reactive. All active oxygen species can be generated by the incomplete reduction of O2 during respiration or by other means. It has been shown that two modes of killing by H2O2 exist in E. coli. Mode one killing appears to be due to DNA damage, reaches it's maximum near 1 to 3 mM H2O2 and requires metabolically active cells during exposure. Mode two exhibits a classical dose-response curve and occurs in the absence of metabolism. The location of lethal cell damage has not been identified for this mode of killing. These is no evidence about the killing patterns induced by H2O2 in Salmonella typhimurium. In our experiments the modes of killing induced by H2O2 in S. typhimurium were investigeted. We treated strains G46, TA1950 and TA104 in different phases of bycterial growth with various concentrations of H2O2. We also tried to find out the possible correlation betewwn the killing rate and mutagenesis. In comparison with data obtained with E. coli, our results show the best differences in the mode of killing between these3 two species of bacteria when exposed to the same agent. The results of mutagenesis show the differences among the strains according to their genotypes.
