MECHANICAL TESTING OF THE BONES OF AGED OVARIECTOMIZED RATS

Testing of bones is applied in medical diagnostic for evaluation of their mechanical characteristics. Sevelamer hydrochloride (Renagel® ), a non-calcium phosphate binder, has been shown to reduce coronary artery and aortic calcification and increase trabecular bone mineral density in patients with chronic kidney disease as compared to calcium containing phosphate binders. Here we tested whether sevelamer-HCl affects biomechanical characteristics in aged ovariectomized (OVX) rats when given orally for 12 weeks with normal food. The femurs were removed at death rats, prepared for biomechanical analysis. All experiments and protocols were approved by the Institutional Animal Care Committee. Using a materials testing system (TA.HDplus Texture Analyser, The Stable Micro Systems) with 30 kg load cell, two types of mechanical testing were performed on the femur: three-point bending test and indentation test. The three-point bending results show that maximal load and stiffness were 17% and 15% higher in sevelamer treated animals as compared to OVX rats. The indentation test results show that the direct parameters: maximal load, stiffness and energy absorbed were significantly increased in sevelamer treated rats as compared to OVX control animals ; the ultimate strength showed the same trend. Sevelamer treatment significantly improved the trabecular bone parameters as compared to OVX animals, but did not restore the mechanical properties to those exhibited by sham rats.
