The effect of the substrate water content on the ochratoxin A production by mould Aspergillus ochraceus NRRL 3174 in the pure culture
The aim of this study was to determine the water content in a solid substrate - corn, optimal for the pure culture of the mould A. ochraceus NRRL 3174 biomass growth and for the highest production of ochratoxin A (OTA). The growth of the mould investigated, as well as the OTA biosynthesis, were followed during the 35 days of incubation in a stacionary culture, at temperatures 20 oC and 28 oC, on corn grains with initial water content of 28% or 38%. Biomass synthesis was determined indirectly by measuring chitin, and OTA concentrations were measured by fluorodensitometer. The results have shown: (a) during the cultivation at 20 oC and 28% water content in the substrate, the maximal biomass synthesis (4.20 mg x g-1 of substarte) was reached after 14 days, with the peak of OTA production (16.50 mg x g-1 dry weight mycelium). Under the same conditions, but with 38% water content in the substrate, the biomass growth and the OTA production were 30% and 20% higher, respectively. (b) the cultivation at 28 oC gave similar results. On the substrate with 28% water content the highest OTA concentration (14.20 mg x g-1 dry weight mycelium) was measured after 14 days of incubation, and was about 15% less than under the same conditions on the substrate with 38% water content. The biomass growth on the substarte with 28% water content reached the maximum after 35 days, with the values 15% less when compared with the growth on the substrate with 38% water content. According to the results obtained for OTA production and for the biomass synthesis of the mould A. ochraceus NRRL 3174 grown in the pure culture under the certain parameters, it was concluded that 20 oC and 38% water content in the substrate were optimal for the growth of biomass which produced the highest amount of OTA.
