Rapid LC-NMR and LC-MS characterization of an unknown component in a novel antifungal drug Icofungipen

A successful use of LC-NMR and LC-MS for a rapid identification of the unknown component in a novel antifungal drug Icofungipen has been demonstrated. Complementary information obtained from the two methods made it possible to identify and determine the structure of A1 prior to its isolation and purification. The analysis of the stop-flow LC-NMR (1H and DQFCOSY), LC-MS and LC-MS/MS spectra has shown that A1 is structurally related to Icofungipen. The impurity A1 was later isolated and prepared synthetically and its structure was corroborated by high-resolution NMR spectroscopy. The combined use of LC-NMR and LC-MS methodology as described here can serve as an efficient, fast and reliable tool for determination of the Icofungipen impurities in the following production process.
