Cell cultivation and biomass production: BHK-21 C13 cells in toxicology and vaccine production

BHK-21 C13 cell line in suspension culture is used as a model. The present results are compared with the aggregates grown in stirred vessels with different working volumes and mixing conditions.Cell cultivation with initial cell bank (ATCC) and expansion to final scale fermentation is carried out with specifically designed culture medium. Scale up of the BHK-derived cells starts with an initial cultivation at low cell density in tissue flasks and cell-spin bioreactors. Bioreactor BR-06 equiped with electronically controlled four vertical pole pieces producing rotating magnetic field and automatic oxygen and pH control was used. Cells were cultivated using batch or semi-continous batch procedure. Growth behaviour of BHK-21 C13 cells grown in suspension was maximal growth rate mmax = 0.032, viability 92-98%, population doubling time t_d/h 16.14 and maximum cell density 1.48x10^6 cells/ml. The harmful effects of chemicals on ecosystem are connected with their entry, distribution, bioaccumulation, binding and degradation. The quantitative determination of the effects of xenobiotics or toxins on whole organisms and on cellular function is better done under laboratory conditions. BHK cells are particulary useful for application in virusology.
