Atrazine toxicity on the rat prostate subcellular mechanisms
Interaction between ^3H-5alpha-dihydrotestosterone and its specific nuclear receptors (NR) in rat prostate (P) under atrazine (A) influences were studied. Scatchard plots for specific binding revealed K_d value 2.5 x 10^-12 M and binding sites" of 422 fmol/mg of DNA. In vitro 0.7 micromol of A induces decrease in number of binding sites on NR molecules (286 fmol/mg of DNA), while K_d remains in the same range. Protein contents in P cytosol were followed in vivo treatment per os with 8 or 12 mg A per kg b. w./10 days. Proteins in P cytosol were resolved by 1-D 12% SDS PAGE after Commassie blue staining and quantified by UltroScan XL densitometer. The decreases from 6 to 44% in protein contents at Mr 10.5 kD, 12.8 kD, 15.1 kD, 21.7 kD, 31.6 kD, 50.9 kD and 62.3 kD were detected vs. controls. The highest changes in protein band contents were detected at Mr 31.6 kD and 50.9 kD in samples from the treated rats with the higher dose. The study present findings in subcellular changes under A influence at rat P level, which is one of the most widely used experimental systems for studying the androgen gonadotropic feedback regulation.
