The induction of the aidB gene from Escherichia coli in an ada-independent fashion and by alkylating agents in Salmonella typhimurium

The E. coli cells have the ability to acquire increased resistance to the lethal and mutagenic effects of alkylating agents during exposure of cells to sublethal doses of methylating and ethylating agents. This phenomenon has been termed the adaptive response and requires a functional ada gene. In contrast to the case with E. coli, the adaptive response does not seem to occur in the closely related species S. typhimurium. The unique feature of the aidB gene, as a member of adaptive response, is that it can be induced by anaerobiosis and cytoplsmic acidification in an ada- and alkylation- independent fashion. Our interest was to investigate the influences of oxygen-limiting conditions and changes in intracellular pH on the expression of adiB in E. coli. Induction of aidB gene was monitored by assaying ß-galactosidase activity in extracts obtained from strains containing operon fusion of Mud1 (bla lac) to aidB. We obseved that both anaerobiosis and supplementing acetate to the growth medium at a pH range of 6.0 to 6.8 resulted in aidB activation. The induction of aidB gene as the aidB::lacZ fusion in S. typhimurium was tested by N-methyl-N"-nitro-N-nitrosoguanidine, methyl methanosulfate, N-methyl-N-nitrosourea and ethyl methanosulfate treatment. it was also of interest to compare the induction of the aidB gene with the induction of the SOS response. We showed that treatment with alkylating agents lead zto decrease in the expression of the aidB gene when the uvrB mutation was present.
