Functional role of S-layer protein from probiotic strain Lactobacillus helveticus M92
The objective of this study was characterisation of the S-layer protein (SlpA) and its role in probiotic activity of Lactobacillus helveticus M92. SlpA was isolated, purified and identified by SDS-PAGE LC-MS/MS analysis. SlpA gene from L. helveticus M92 is sequenced and compared with other sequenced S-layer protein genes of Lactobacillus strains by BLAST pairwise alignments. Protective function of S-layer protein during preparation of dry probiotic culture by lyophilisation was observed. Furthermore, the purified S-layer proteins has shown better immunomodulatory effect than L. helveticus M92 cells after S-layer protein removal in orally immunised mice. The possible role of S-layer protein in competitive exclusion of Salmonella enterica serovar Typhimurium was also examined. The coagregation of L. helveticus M92 with S. Typhimurium was 10, 52 % lower when S-layer proteins were removed from the cell surface. Therefore, the influence of the oral treatment with probiotic strain Lactobacillus helveticus M92 on an oral infection of mice by S. Typhimurium was investigated. After this treatment, the total number of lactic acid bacteria in faeces and in intestinal homogenates of mice was increased, contrary, the total number of enterobacteria and Salmonella sp. was reduced. Following the oral immunization of mice, with viable L. helveticus M92 and S. Typhimurium cells, the concentration of luminal content of total S-IgA and specific anti-Salmonella S-IgA antibodies, from all immunized mice was significantly higher compared to the control group or group of mice infected only with S. Typhimurium. These results demonstrate that reduced infection by S. Typhimurim in mice with L. helveticus M92 is associated with competitive exclusion in the intestinal tract and enhanced immune protection conferred by the L. helveticus M92 and its S-layer protein.
