Semi-solid state saccharification and fermentation of corn grits to lactic acid by amylolytic lactic acid bacterium

Utilization of relatively cheap and abundant raw materials in efficient semi-solid state saccharification and fermentation (SSSF) to lactic acid by amylolytic lactic acid bacterium (ALAB) was investigated. Corn grits (initial concentration of 110 g/L) in distilled water was used as a minimal medium for production of lactic acid by homofermentative ALAB Lactobacillus amylovorus DSM 20531T in a lab-scale stirred tank reactor. Efficiency of the SSSF was compared to the efficiency of bioprocesses carried out in MRS medium with corn grits (1) and MRS medium with soluble starch (2). During the bioprocess a portion of semi-solid phase decreased from 34% to 9%, while the concentration of insoluble substrate was reduced to 8% of its initial value. Insoluble starch from corn grits (semi-solid phase) was hydrolyzed and solubilized (liquid phase), and 37% of soluble starch was fermented to d/l-lactic acid mixture (0.66:0.33). Values of YP/S were 0.91 g/g for (1), 0.90 g/g for (2), and 0.71 g/g for the SSSF. The results obtained clearly show the potential of this new bioprocess and its implementation in the production of lactic acid- derived polymers and similar emerging technologies.
