Decrease of bovine acetylcholinesterase activity after the exposure to triazole pesticides
Triazole fugicides Orius 25EW® and Prosaro 250EC® (active substances: tebuconazole and respective tebuconazole plus prothioconazole)are used for the protection of wheat, barley and cole-seed against fungal diseases. Their fungicide effect is a result the inhibition of lanosterol 14-α-demetylase (CYP 51). The enzyme is included in ergosterol formation, an essential component of fungal cell membrane. Decrease of acetylcholinesterase (AChE) represents one of the important environmental contamination parameters. Inhibition of AChE causes the acetylcholine accumulation in synapses and neuromuscular plates and subsequent hyperactivation of brain and muscle tissue. AChE is localized in erythrocytes, synapses, neuromuscular plate and heart. Experiments were performed with bovine erythrocytes as a source of AChE. Changes in acetylcholinesterase activity were assessed spectrophotometrically after the exposure to Orius 25EW® and Prosaro 250EC® in vitro at concentrations of 50, 100, 150, 200, 250 and 300 µg.ml-1. Changes in absorbance were read and statistically evaluated by linear regression model (GraphPadPrism). Our results confirmed a high statistically significant inhibition of AChE activity (p < 0.001) in bovine erythrocytes exposed to both fungicides tested.
