NMR Methods in Unravelling Macrolide Antibiotics Interactions
One and two-dimensional NMR techniques have proven usefull in conformational analysis and structure-based inhibotor design. We have shown that a systematic approach which combined one- and two-dimensional NMR methods and molecular modelling calculation could generally be applicable to conformational studies of macrolides free and bound to ribosomes. Furthermore, an application of NOE based experiments such as saturation transfer difference (STD) provides further information on the bound state conformation and binding epitopes. The application of paramagnetic relaxation enhancement (PRE) induced by an inert lanthanide complex can reveal the immersion depth of macrolide antibiotics bound to their targets. In this talk the interactions of macrolides with ribosomes, membrane mimetics and bile acids will be presented and discussed. The knowledge gained from these studies can serve as a platform for the design of novel compounds with an improved biological profile.
