Composition of non-structural and structural carbohydrates in plant secondary raw materials

Reutilization of agro-industrial waste as secondary raw material in functional food production is gaining a significant interest in recent years due to their nutritional value and many economical and ecological arguments. Among other components, carbohydrates are the most abundant, present as non-structural (soluble sugars) and complex structural fractions i.e. dietary fibers. Compositional determination of the latter may indicate the potential for their reprocessing into oligosaccharides with prebiotic properties. Therefore, the aim of this research was to evaluate the content and composition of non-structural and structural carbohydrates in cocoa husks (CH), walnut shells (WS) and pea pods (PP), materials not well described in literature in the given context. Non-structural sugars were recovered using repeated extractions in 50% MeOH solution and analysed using HPLC-RID. The content of structural carbohydrates was evaluated as fibre content using AOAC 2011.25 procedure, while their composition by subjecting the extracted residue to acid hydrolysis and afterward HPLC-UV analysis coupled with pre-column derivatization using 3-methyl-1-phenly-2-pyrazoline-5-one (PMP). Although almost negligible in CH and WS (less than 0.2% DM), soluble sugars comprised a substantial content in PP (10.38% DM). Fibre analysis revealed the predominant content of the insoluble fraction of more than 50% DM, with WS as the richest source (91% DM), which may be attributed to relatively high lignin content. Qualitative analysis of structural carbohydrates showed the presence of galacturonic acid, galactose, glucose, mannose, xylose, arabinose and rhamnose in different proportions, with total glucan fraction being the most represented, followed by total xylan fraction in WS and PP samples and galacturonan fraction in CH. The obtained results demonstrated the carbohydrate variety in secondary raw materials by means of content and composition. Relatively high xylan and galacturonan fraction in the evaluated substrates may present a potential for hemicellulose and pectin reutilization as functional xylooligosaccharides and pectic oligosaccharides.
