Minimal artificial enzymes: development of a new class of chiral catalysts for asymmetric reactions
The development of many small-molecule catalysts is inspired by nature, for example by metalloenzyme active sites. However, despite the progress in asymmetric catalysis with small molecule transition metal catalysts, they are still easily outperformed by metalloenzymes in terms of both selectivity and reaction rates. Obviously, mimicking the active site alone is not sufficient to replicate metalloenzyme activity. The major difference between enzyme active site mimics and metalloenzymes is the outer-coordination sphere present in the later. The basic idea of our work is to design artificial metalloenzymes that contain not only the active site but also a minimal but functional outer-coordination sphere. The outer-coordination sphere should be minimal in order to retain favorable properties of small-molecule catalysts, namely cheap synthesis by chemical methods as well as potential application to a broad range of substrates. The function of the outer-coordination sphere of the proposed catalysts is transmission of chirality. In asymmetric transition metal catalysis it is widely accepted that the chiral information should be incorporated as close as possible to the coordination sphere of the catalytically active metal centre. Within our work we challenge this important principle and use a catalytic metal that is only prochiral. The chiral information is transmitted by "backdoor induction" via hydrogen bonding of distant pendant amino acids or short peptides. In this communication, selected examples of minimal artificial enzymes will be presented.
