Determination of histamine in fish by Surface Enhanced Raman Spectroscopy – comparison with reference HPLC method

Surface enhanced Raman spectroscopy (SERS) method for histamine determination in fish samples using silver colloid SERS substrates was previously developed. This work was conducted in order to test the SERS method on real samples and compare the results with reference HPLC method. Atlantic mackerel samples were divided in 3 groups – fresh and stored at 25°C for 8 and 24 h. Fresh samples were spiked with different histamine concentrations and calibration curves based on intensity of histamine SERS band at 1264 and 1570 cm-1 were constructed by linear regression with R2 values of 0.9849 and 0.9028, respectively. Since model based on intensity of histamine SERS band at 1264 cm-1 performed better, it was used for determination of histamine content in fish samples. Three groups of samples were subsequently analyzed by SERS and HPLC method and obtained results were compared. Average recovery of the SERS method in histamine concentration range 0 – 200 mg/kg was 104.3 % with relative standard deviations of 9.86%. HPLC analysis of fish samples showed that second group of samples contained no histamine, while average value of histamine content in third group of samples was 29.73 mg/kg with RSD of 0.46%. Average value of histamine content obtained by SERS method was 1.62 mg/kg for second group and 28.33 mg/kg for third group of samples with RSD of 12.81%. Trueness of SERS method in relation to HPLC method was 95.31%.
