Photosynthetic purple non-sulphur bacteria as a potential 5-aminolevulinic acid producers

5 aminolevulinic acid (5-ALA) can be used as an effective and ecological herbicide, plant stress tolerance enhancer, tumor-localizing and photodynamic therapy agent. In this research, different strains of purple non-sulphur bacteria from genera Rhodobacter were isolated and, afterwards, genetically modified to improve 5-ALA production. Obtained mutant strains were used to define the optimal bioprocess conditions for growth and 5-ALA production in Erlenmeyer flasks (EF) and stirred tank bioreactor (STB). Rhodobacter capsulatus isolates were genetically modified by N-methyl-N-nitro-N- nitrosoguanidine and high 5-ALA producing strain was selected. Cultivation of mutant strain R. capsulatus B-6508 and 5-ALA production in different conditions (different carbon sources, light/dark regime, aerobic/microaerophilic conditions) was carried out. Samples were analysed by standard microbiological methods and supernatant composition was determined by GC-FID method. 5-ALA concentration of 579 mg/L was achieved with addition of Gly and succinate of 40 mM, levulinic acid 15 mM and glucose concentration of 4 g/L, under microaerophilic light conditions in EF what is 53 times higher compared to the wild strain. This mutant strain under dark aerobic conditions in the STB produced 290 mg/L of 5-ALA. Cultivation of R. capsulatus B-6508 in dark aerobic conditions resulted in satisfy 5-ALA production in the STB, but further improve of 5-ALA production can be achieved by using different cultivation techniques such as fed- batch, repeated batch or continuous techniques.
