Isolation and structure of flavonoids from peppermint (Mentha x piperita) leaves
Mentha x piperita L. (Lamiaceae) has been known for a long as a medical plant, and its therapeutic effects were mostly attributed to the composition of its essential oil. In addition to the essential oil, it is known that M. x piperita contains some other pharmacologically active ingredients as well, such as tannins, bitter substances, caffeic acid and flavonoids. The biological activity of these substances, can become prominent when the herb or its extracts are administered in the form of teas or medicinal preparations. In this study, flavonoids were isolated from the test material by various methods. It should be noted that here TLC was not used for purifying flavonoids already separated but as direct isolation and separation process. The structure of major isolated flavonoids was determined by using combined methods of IR and UV/VIS spectrometry, mass spectrometry and thin-layer chromatography. The thin-layer chromatography on microcrystalline cellulose proved to be an efficient and simple method for isolating and separating flavonoids of peppermint. The purity of the obtained flavonoids was sufficient for direct structure analysis. This study confirmed the presence of luteolin-7-O-rutinoside and eriodictyol-7-O-rutinoside as major flavonoids in the examined plant material cultivated in Eastern Slavonia.
