Glycosylation in stress and disease

Oligosaccharides have a whole spectrum of biological roles, from those that appear trivial, to those which are crucial for the development, growth, defense and function of survival of the organism. By reviewing recent evidence of the importance of glycosylation, which seems to be the only three-dimensional posttranslational protein modification (112 references), we focus on the role of oligosaccharides in intracellular trafficking, cell-cell and cell-matrix interactions, selectin-oligosaccharides interactions in leukocyte adhesion, changed protein glycosylation in stress and in developing acute illness, such as septic shock, allergies, rheumatoid arthritis, or neoplastic diseases. Diagnostic relevance of modified glycosylation in plasma proteins and tumor products is discussed pointing to glycoproteins as emerging candidates for potent tools in differential diagnosis. The accumulation of knowledge concerning the importance of glycosylation is accelerating exponentially. Nevertheless, the specific role of given oligosaccharide structure is still impossible to predict thus keeping glycobiology on the hot frontier of today’s life sciences.
