Synthesis of sulfamethoxazole metabolites

Sulfamethoxazole (SMX) is a well known antibacterial drug. Most serious side effects of sulfamethoxazole and other sulfonamides are hypersensitivity reactions, the pathogenesis of which has been suggested to be mediated by reactive metabolites. We report here the new synthetic paths for preparation of sulfamethoxazole: 4-nitrozo-N-(5-methy-3-isoyazolyl)benzenesulfonamide (SMX-NO), 4-nitro-N-(5-methy-3-isoxazolyl) benzenesulfonamide (SMX-NO2), azoxybenzene-4,4-(N-5-methy-3-isoxazolyl)disulfonamide (SMX-AZ), N-(4-aminobenzenesulfoniyl)-3-oxo-butyramidine (SMX-AD) and 4-hydroxylamino-N-(5-methyl-3-isoxazolyl)benzenesulfonamide (SMX-HA). Such compounds may be useful in the development of rapid, accurate diagnostic tests for hypersensitivity of sulfamethoxazole.
