Liposomes with capsacin, chili extract and chili powder: characterization

Topical creams with capsaicin are used to treat post herpetic neuralgia and diabetic neuropathy, osteoarthritis and rheumatoid arthritis. Capsaicin has also been used to treat pain de to pruritus, psoriasis, mastectomy, bladder disorders and cluster headaches. Adverse (unwanted) events from capsaicin are mainly at the application site (burning, stinging, erythema), and systemic events are extremely rare. Respiratory irritation has also been reported from inhalation of dried cream. In order to increase the efficacy of capsaicin delivery into the skin and to reduce the possible side effects (by reducing the dose necessary to achieve the effect), the entrapment of capsaicin in liposomes, vesicles with proven superiority in dermatology, was suggested. By incorporating capsaicin in liposomes, it is expected that the penetration of capsaicin into skin will increase and that side effects will be minimized. Liposomal suspensions are also preferable as a vehicle for the skin application, since both alcoholic solutions (like in Capsicum tincture) and oily extracts (like in Ayurvedic preparations) are with the limitations regarding their spreadability problems, staining the patient’ s cloths, etc. The main objectives of our research were to optimise the entrapment of Capsicum (in the form of pure capsaicin, Capsicum tincture or crude chilli powder) in liposomes, to characterize the liposomal suspensions and to perform in vitro release studies. The phospholipid composition and liposome preparation methods were varied in order to optimise the preparation. Liposomal suspensions were characterized for particle size and the results will be presented.
