Epigenetic modulation of the cell membrane N-glycome

Membranes of all cells are covered with a dense layer of complex oligosaccharides (glycans) which represents an essential element of all interactions between the cell and the environment. The vast majority of pathogens use glycans to attach to target cell, while the cell itself regulates adaptive properties of the membrane by modulating membrane protein glycosylation. However, contrary to proteins which are defined by sequence of nucleotides in individual genes, glycans are encoded in a dynamic network encompassing thousands of different proteins which participate in the complex process of protein glycosylation. In addition to genetic polymorphisms in all these genes, the exact structures of produced glycans are affected by various past and concurrent events in the cells which affect activities of numerous relevant proteins. Epigenetic regulation of gene expression, as one of the major mechanisms how cell adapts to environmental changes, is a prominent mechanism in the process of glycosylation as well. Nevertheless, studies of epigenetic regulation of glycosylation are still very limited. Aiming to investigate effects of major epigenetic inhibitors on the cell membrane glycome, we analyzed effects of zebularine, 5-aza- 2’-deoxycytidine, Trichostatine A and Na-butyrate on N-glycome of HeLa cell membrane. Significant changes of the membrane glycome were observed after treatment with each of the epigenetic inhibitors ; a number of different glycans was upregulated, while others were downregulated. While some effects were common for different inhibitors, some effects were specific for individual inhibitors. Membrane glycans are the point of attachment for numerous pathogens and with the increasing number of epigenetic inhibitors being registered as drugs, this observation suggest that they might also be used as anti-adhesive microbial drugs in some specific situations.
