Vaginal penetration of chitosan-coated liposomes with clotrimazole: the effect of vesicle size
Phosphatidylcholine liposomes containing clotrimazole were prepared by the modified film method and sonicated, when applicable, to desired vesicle size. Low molecular weight chitosan was used as coating polymer. Determination of vesicle size and size distributions of uncoated and coated vesicles showed that coated vesicles are bigger in size, as expected. In addition, we could detect the influence of polymer concentration on vesicle size ; vesicles with higher concentration of polymer (0.6 % ; w/w) being larger in size than those with lower concentration (0.1 % ; w/w). Clotrimazole entrapment was also influenced by the coating process and chitosan concentration. Drug release determined on Franz diffusion cells, both through artificial membrane and vaginal tissue, indicated that vesicle size and chitosan coating affect the drug release, wherein the coated liposomes demonstrated slower release of clotrimazole. Mucoadhesivness, as evaluated on vaginal tissue, was affected by polymer concentration used for vesicle coating, as well as vesicle size.
