Characterization and in vitro evaluation of deformable liposomes for vaginal delivery
Deformable liposomes composed of egg phosphatidylcholine (85%) and sodium deoxycholate (15%) containing metronidazole or clotrimazole were prepared by the modified ethanol injection method. Physicochemical characterization of various liposomes was based on determining the size, the polidispersity, zeta potential and entrapment efficiency of drugs. The optimal formulations of deformable vesicles containing either metronidazole or clotrimazole, were incorporated in hydrogel composed of 1% Carbopol 974P NF. In vitro release studies of the drugs entrapped in liposomes and incorporated in gels performed in the buffer pH 4.5 and vaginal fluid simulant pH 4.5 have demonstrated sustained release of both, clotrimazole and metronidazole. Components of vaginal fluid and lower pH (pH 4.5) influenced the stability of liposomes, as compared to control study (pH 6). Regarding the physical properties of liposomal gels, different rheological measurements and mucoadhesiveness on vaginal tissue were performed and will be discussed.
