Glycosylation of immunoglobulin G in menstrual cycle

Specific immunoglobulin G (IgG) glycoforms are being evaluated as biomarkers for different diseases (tumors, autoimmune diseases etc.) regarding disease predisposition, prognosis and answer to therapy. However, IgG glycosylation also changes with various physiological parameters, such as age and sex. It is also known to change in pregnancy and under influence of estrogen. In order to be certain that changes in IgG glycosylation pattern are indeed associated with a disease when it comes to the population of pre-menopausal women, it is important to investigate whether normal hormonally governed physiological processes, such as menstrual cycle, influence IgG glycosylation. The aim of this study was therefore to follow IgG glycosylation patterns during menstrual cycle. The study was conducted on female individuals with regular menstrual cycle, non-smoking and non- drinking history that were not using oral hormonal contraceptives. Once a week 5 ml of peripheral blood was drawn and plasma was separated. IgG was isolated from plasma samples by affinity chromatography on a protein G monolithic plate and its N glycans were released by PNGase F, labeled with 2 aminobenzamide and analyzed by hydrophilic interaction ultra-performance liquid chromatography. Using this approach, we performed a longitudinal comparison of IgG glycosylation profile for each female individual in different stages of menstrual cycle during a period of 12 weeks.
