Lamotrigine and modulation of Na+, K+-ATPase activity in rat hippocampus and cortex after kainic acid induced seizures

Na+, K+-ATPase is the enzyme responsible for maintaining the ionic gradient necessary for normal neuronal excitability. We examined the effects of kainic acid (KA) induced seizures on the activity of Na+, K+-ATPase in the rat hippocampus and cortex. We also investigated the influence of lamotrigine (LTG) pretreatment on the modulation of the enzyme activity. The enzyme activity from hippocampal and cortex tissue was determined two hours and three and five days after a single subcutaneous KA (8 mg kg) injection. The activity of Na+, K+-ATPase was also determined in rats pretreated with LTG (30 mg/kg) two hours before KA application. The Na+, K+-ATPase activity in the hippocampus was significantly inhibited the third and fifth day after KA application. In the group of the LTG- pretreated rats the Na+, K+-ATPase inhibition was detected the fifth day after KA treatment. The inhibition of Na+, K+-ATPase observed the fifth day after KA administration was significantly less pronounced in the LTG- pretreated rats. The enzyme activity in the cortex of KA- treated rats was significantly inhibited the fifth day. In the LTG- pretreated animals there was no significant inhibition of the cortical Na+, K+-ATPase activity. It may be concluded that prolonged convulsions caused by KA application showed significant inhibition of the Na+, K+-ATPase activity in the rat hippocampus and cortex. LTG showed a partially protective effect in the hippocampus of KA- treated rats and a completely protective effect in the cortex.
