Antiproliferative and Antioxidant Activities of the Organic Extract and Semi-purified Fractions from Gorgonian Coral Eunicella cavolini

The yellow gorgonian, Eunicella cavolini (Koch, 1887), is a common and widespread gorgonian species in the Mediterranean Sea known as an abundant source of structurally different primary and secondary metabolites [1]. In a large number of studies on gorgonians, diterpenes, sesquiterpenes, alkaloids and steroids, often unique in carbon skeleton and stereochemistry were isolated and characterized. Many of these compounds have shown a wide range of biological activities, which has attracted the attention of the scientific community, particularly in the search for new anticancer compounds of natural origin [2–4]. Within the platform of Adriatic Sea bioprospecting, we evaluated the antiproliferative and antioxidant activities of the organic extract and corresponding, four semi-purified fractions from E. cavolini. The organic extract was obtained by using dichloromethane/methanol solvent system (3:1) followed by fractionation using a solvent system (cyclohexane, ethyl acetate, methanol) of different ratios. The antiproliferative activity was assayed against four human cancer cell lines (SW620, CFPAC-1, MCF-7 and HeLa) as well as non- transformed fibroblast cell line (HFF-1) using MTT [3-(4, 5- dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide] colorimetric method. The antioxidant activity was examined by 1, 1- diphenyl-2-picrylhydrazyl (DPPH) assay. All tested samples displayed strong antiproliferative activity with IC50 value around or less than 50 μg/mL, except for the first fraction, F1 (c-hexane/ethyl acetate, 75:25) against SW620 colon cancer cells. Fraction, F3 (ethyl acetate) exhibited the strongest radical scavenging activity with IC50 value of 0.76 mg/mL. Further studies will include the isolation of pure bioactive secondary metabolites and the assignation of their structures by NMR spectroscopy.
