Histochemica investigations of nadph- and nadh-tetrazolium reductase activity in the liver of selenium and copper treated carp (Cyprinus carpio L.)

Histochemical methods were employed to examine the effects of selenium and copper intoxication on NADPH- or NADH-tetrazolium reductase activity in liver tissue. Both enzymes were located in the intracellular granules though NADH-tetrazolium reductase also showed diffuse distribution in the cytoplasm. The granules in which the activity of both enzymes was demonstrated represent mitochondria, while the activity of NADH-TR in the cytoplasm refers to the endoplasmic reticulum. The results showed that selenium and copper in concentrations of 0.1 mg/L did not induce changes in NADPH-TR activity. The activity of NADH-TR in the mitochondria oscillated throughout the treatment period with both elements, while copper also induced a significant increase of enzyme activity in the cytoplasm. Prolonged intoxication with both selenium and copper lead to damages in liver tissue and the reduction of activity for both enzymes.
