Quinoline fumardiamides as novel quorum sensing inibitors
Conventional antibiotics are inefficient in the treatment of biofilm-associated infections, which complicate treatment of various infections.1 In biofilms, where cells exist as heterogeneous populations with various growth rates (including dormant cells), embedded in a protective matrix, cell-to-cell communication plays an important role.2 This process, termed quorum sensing (QS), seems to be crucial for biofilm formation, and it has emerged as a prominent target for finding effective strategies aimed at biofilm control. In our previous work we have shown that asymmetric primaquine and halogenaniline fumardiamides (1a-f) showed significant activity in biofilm eradication assay.3 As a continuation of our research, we have designed and prepared analogous derivatives with mefloquine (2a-f) and chloroquine (3a-f) pharmacophores and tested anti-QS and bactericidal activity of all quinoline derivatives 1-3.
