Probiotic activity of Bifidobacterium animalis subsp. lactis BB-12® in dehydrated wheat/rice cereal products

Probiotic activity of Bifidobacterium animalis subsp. lactis BB-12® in dehydrated wheat/rice cereal products developed for infants older than 4 months was investigated. The number of live BB-12® cells in three samples was above the minimal required number of probiotic cells per gram of product (1x106 CFU/g) till the end of the product expiration dates. Significantly higher viable cell counts were obtained in simulated gastrointestinal tract conditions when probiotic culture BB-12® was applied in processed wheat/rice cereal products compared to cell count of pure lyophilized culture. Also, wheat/rice cereal matrices contributed to the BB-12® in vitro cholesterol assimilation ability and antibacterial activity. Adhesion properties of BB-12® strain to Caco-2 cell line and proteins of extracellular matrix and competitive exclusion of test-microorganisms were investigated, whereas adhesion of BB-12® strain to Caco-2 cell monolayer was measured by fluorescence microscopy. Among conditions tested (MOI= 10 ; 100, incubation time 1h, 4h, 12h), the highest ratio of 35 BB-12® cells per 100 Caco-2 cells, was obtained at MOI 100 and after 12 hours of incubation. Additionally, the occurrence of BB-12® strain adhered to Caco-2 cell line reduced the attachment of S. Typhimurium FP1 for approximately 1.5 log10 CFU/ml, and of E. coli 3014 for approximately 0.71 log10 CFU/ml. Hence, considering the beforehand gained adhesion pattern, proved by live cell imaging microscope together with exclusion capacity, adhesion to extracellular matrix proteins and coaggregation with examined pathogens, BB-12® probiotic strain has an impact on the adhesion ability and could downsize the negative effects of both strains, S. Typhimurium FP1 and E. coli 3014.
