Influence of Acute and Intermittent Hyperbaric Oxygenation on Oxidative Stress Level and Reactivity of Middle Cerebral Artery in Healthy Sprague-Dawley Rats

Introduction: Hyperbaric oxygenation (HBO2) has been widely accepted adjuvant therapy for various conditions with impaired tissue oxygenation, such as ischemic injuries, diabetic foot and ulcers, sepsis and skin flap transplantations. Recently, we have demonstrated beneficiary effects of HBO2 on brain infarct volume in the model of diabetic female Sprague-Dawley rats. Besides its beneficiary effects on the tissue perfusion, HBO2 exhibits high toxicity at higher pressures, due to increased oxidative stress and barotrauma. The aim of the present study was to determine if vasoreactivity of cerebral resistance vessels (middle cerebral artery ; MCA) is affected by HBO2, by single or multiple exposures and if potentially observed changes are related to the changes of the oxidative stress levels in the vascular tissue. Materials and Methods: Male Sprague-Dawley rats were divided in control, single exposure HBO2group (AHBO2) and 4-days consecutive exposure to HBO2 group(4D-HBO2). The following day after HBO2 treatment, rats were sacrificed by decapitation under anesthesia and analgesia and MCA were isolated for in vitro vasorectivity studies. Cerebral blood vessels were harvested for antioxidative enzymes’ protein expression analysis as well as blood sample were taken for the markers of systemic oxidative stress. Results: HBO2 significantly affected flow- induced dilation of MCA. AHBO2 significantly affects antioxidative enzymes’ expression (such as SOD1-3 isoforms, and gluthatione-peroxidase isoforms), but not uniformly. Conclusion: Study confirms the hypothesis that HBO2 exposure, depending of protocol may change the level of oxidative stress which may be underlying observed changes in vasodilation of cerebral resistance vessels.
