Biologically intriguing N-sulfonyl derivatives of pyrimidine nucleobases and their metal complexes

The crystal structure of the recently synthesised ligand, N-1-sulfonylcytosine derivative (1), as well as synthesis, spectroscopic and structural studies of its Cu (II) (2) and Co(II) (3) complexes are presented. The N-sulfonyl derivatives of pyrimidine nucleobases exhibit strong in vitro antitumor activity. In order to increase its solubility, the metal complexes Cu[1]Cl2 and the Co complex with the putative formula Co[1]Cl2 were prepared by reaction of CuCl2 x 2H2O i.e. CoCl2 x 6H2O with ligand 1 in methanol, using 1:2 metal to ligand ratio. The colour change indicated the complex formation in both cases. The X-ray structure analysis of 1 (Fig. 1) shows the ligand in the keto-amino form whereas a mixed amino-imino form was revealed in the Cu(II) complex 2 (Fig. 2). The expected Ci symmetry of the molecule 2 with the Cu2+ ion at the inversion centre was not observed. The Cu2+ ion lies at the centre of the severely distorted octahedron, coordinated by two pyrimidine nitrogen atoms (N3 and N3') from two ligand molecules, as well as by two keto oxygen atoms (O3 and O3'). Two chlorine atoms complete the six-fold coordination. The overall conformation of the ligand was preserved in its Cu(II) complex. In the structure of 2, two clearly resolved methanol molecules involved into the extensive hydrogen bonding contribute to the overall crystal packing. The sensitive rosy crystals of 3 soon after being taken out of the mother liquor change their colour to blue. However, the tentative unit cell was determined, that confirmed the complex formation. The 1H NMR data suggest that the structure of Co(II) complex 3 is analogous to the Cu(II) complex 2.
