Potential role of CD26 in coeliac disease
Within the last ten years there has been a growing interest in studying exopeptidases that, through their proteolytic activity, regulate large number of physiological and pathological processes. Especially interesting is the CD26 molecule(dipeptidyl-peptidase IV/DPPIV) which is a 110kDa transmembrane glycoprotein.It consists of extracellular, transmembrane and cytoplasmic part.The extracellular part contains of a catalytic domain which cleaves the X-proline dipeptides from the N-terminus of polypeptides. CD26 exists in membrane bound and soluble form.It is found on the surface of epithelial cells of many organs including the gastrointestinal tract, on the surface of lymphocytes, on the capillary endothelial cells, as well as on some parenchymal cells such as hepatocytes.The CD26 molecule has a large number of natural substrates such as cytokines, chemokines and neuropeptides. With its catalytic action, CD26 can activate, inactivate or change the specificity of action of these substrates in both physiological and pathological processes. In addition to its catalytic action, CD26 can regulate physiological and pathological processes by binding with other molecules such as ADA and CD45 on the surface of T-lymphocytes. It has been found that the expression of CD26 is decreased on the surface of epithelial cells of the small intestine in a patient with coeliac disease. Due to this reasons we have decided to study whether the expression of the CD26 is really lowered in patients with coeliac diseases.In future we are planing to induce coeliac disease in CD26+/+ and CD26-/- mouse in order to determine the cause-and-effect relationship between this molecule and this disease.If we succeed in demonstrating that the CD26 is important in the pathogenesis of this disease, we will attempt to determine molecular mechanism by which the CD26 contributes to malnutrition in coeliac disease
