Plant DNA isolation from differently preserved leaf tissue and its use in RAPD analysis
There are many unresolved problems in the taxonomy among and within species of the genus Thalictrum (Ranunculaceae). The answers that are sought could be provided by the application of molecular systematic methods. Varying the DNA isolation techniques, preservation techniques and RAPD with some species of the genus Thalictrum has led to the optimum procedure applicable in taxonomic analysis. The DNA isolation procedure from leaf tissue that uses laundry detergent as a detergent buffer system has been found to be the most suitable. If necessary, DNA isolation from leaf tissue that is preserved at four different ways is possible. For a high quality RAPD analysis, an abridged PCR protocol has been employed.
