Expression cytokeratin 10 and nuclear marker KI67 in keratoacanthoma and squamous cell carcinoma
In our study we compared 50 clinically and histologically diagnosed keratoacanthomas (KA) and 50 squamous cell carcinomas (SCC) with regard to cytokeratin 10 (CK10) and Ki67 expression. CK10 is a high molecular weight keratin that is normally expressed in the suprabasal layer of the epidermis. Ki67 is a monoclonal antibody which detects nuclear antigen present on all actively proliferating cells. In our study percentage of the total stained tumour cells was determined and results were compared. Our results showed higher expression CK10 in KAs than SCCs. A mean od 15-90% of the total area of KAs expressed CK10 with average rate 50.9%. Expression was marked in ordered pattern through the suprabasal layers in 49 out of 50 KAs (98%), whereas a mean of only 2-34% of the total area of KAs expressed Ki67 with average rate 12.4%. Tumour cells KAs showed homogeneous staining of their basal layers with varying degree of intensity. In SCCs a mean of 2-30% of the total tumour area expressed CK10, with average rate 13.06%. Small group of contiguous tumour cells and scattered individual cells expressed CK10 in a disordered pattern. A mean of 12-98% of total tumour area expressed Ki67, with average rate 39.84%. Tumour cells SCCs showed intensely staining within suprabasal layers in 30 out of 50 SCCs (60%). Our results confirm low levels and irregular distribution CK10 expression in SCCs in contrast to KAs where this marker is expressed in a more ordered pattern suggesting a more normal cellular differentiation.
