Optimisation of Separation Amino Acids by Thin-Layer Chromatography
Sequences of 22 different amino acids that are bounded by peptide linkages are specific to each protein, and they can be broken down by hydrolysis. In this paper a possibility of separation and identification of amino acids by thin - layer chromatography was examined. The best separation was achieved on cellulose layer with butanol-acetic acid - water and methanol - chloroform - ammonium (17%) nd by visualisation with the ninhidrine and isatine. A highly sensitive 3CCD colour video camera was used to capture the chromatograms under white light. Optimisation was performed using experimental design software package Design. - Expert 6.
