MICROSATELLITE MARKERS FROM HLA REGION (D6S265, D6S273 AND MIB) IN PATIENTS WITH PSORIATIC ARTHRITIS IN CROATIA

Psoriatic arthritis (PsA) is known to be associated with certain HLA alleles, mainly with HLA class I alleles. The association of HLA with PsA is very complex when the clinical pattern of PsA is compared with HLA distribution. To analyze the genetic contribution of HLA microsatellite loci in development of PsA and to study whether these loci are primary associated with disease or their association is secondary to linkage disequlibrium with HLA genes. Sixty unrelated patients with PsA (33 women and 27 men, mean age 45 years) and 187 healthy controls were examined for 3 highly polymorphic microsatellite loci (D6S265, D6S273, and MIB). The same samples have been typed previously for HLA polymorphism using Polymerase Chain Reaction-Sequence Specific Primers (PCR-SSP) method. Microsatellites were analyzed by electrophoresis on polyacrilamide gel in automated sequencer (ALF express). The results were compared using Chi-square test with Yate’ s correction and the Fischer's exact test. The most frequent alleles at three tested loci among PsA patients were as follows: D6S265-130bp (35.71%), D6S273-132bp (48.98%), and MIB-340bp (14.7%). No significant differences in D6S265, D6S273 and MIB alleles between PsA and controls were found. When we re-evaluated our results after the patients were divided into 2 groups based on the presence or absence of one of HLA-B alleles (B*13, B*27, *39, and *57) associated with PsA in Croatia we observed some differences. Namely, comparison of PsA patients positive for one of HLA-B associated alleles and matched controls showed significantly lower frequency of allele 130bp and 136bp at D6S273 locus, as well as for allele 346bp at MIB locus in PsA patients. It can suggest a protective role of these alleles in developing of PsA. At D6S265 locus we did not observe any difference between patients and controls carrying one of above mentioned HLA-B alleles.
