Efficacious avoidance of CD8+ and NK cells-mediated virus control by MCMV m152 gene product
The sensitivity of some mouse strains (e.g. BALB/c) to MCMV is related to their inability to generate an efficacious NK response during the early post infection period. Here we tested whether the MCMV-sensitive mouse strains are constitutively unable to mount a NK response to this virus or if their sensitivity is due to viral inhibition of NK cell function. We present the evidence that m152/gp40, originally described as a gene that compromise CD8+ T cell function by retaining the MHC class-I complexes in the ERGIC/cis-Golgi compartment, also inhibits NK cell-mediated control of virus growth in vivo. Deletion of m152 abolishes this inhibition rendering otherwise resistant virus-infected cells susceptible to NK cell control. Furthermore, we establish that this NK evasion strategy is ineffectual in resistant mouse strains in part through expression of activating NK cell receptors which are independent of virus-induced inhibitory signals delivered by infected cells.
