On the role of natural killer cell in liver regeneration

Processes involved in the control of normal growth are often studied in the model of liver regeneration, which follows after partial hepatectomy (pHx) or after toxic damage of the liver, but the list of factors acting as primers, progression factors and inhibitors during this growth is still incomplete. Owing to the central role of the lymphatic system in the cytokine network, and its capacity to differentiate between the self and nonself it was hypothesized that natural killer cells (NK) and intermediate NK 1. T cells, as cells of innate immunity and primitive T cells, respectively, might have an important regulatory activity in the control of liver growth. Namely, a number of evidence emphasizes the activation of extrathymic pathways of T-cell differentiation in the regenerating liver, while the current concepts of recognition mechanism point to the ability of these cells to monitor target cells for reduced or aberrant expression of "self" major histocompatibility complex (MHC) class I molecules and react on absence or structural changes of self antigens. The influence of MHC class I expression on liver regeneration is illustrated by data, showing the initial decline (at 24h) and subsequent increase (at 48h and 7 days after pHx) of the proportion of hepatocytes in S phase cell cycle in regenerating liver of mice homozygous for b2-m-/- genotype, which do not express correctly folded MHC class I trimolecular complexes and lack CD8+ T cells, and by the data showing that in vivo depletion of NK1.1.+ cells increases the early phase of liver regeneration in heterozygous b2-m+/- mice. The significance of the appearance of autoreactive clones, confined mostly to NK1.1.+ CD4- CD8- IL-2Rb+ T cells expressing intermediate level of the T cell receptor was also discussed.
