It is often emphasized that biliary obstruction induces a significant immune dysfunction, which may be responsible for perioperative complications and high mortality rate (even to 13%.) The effects might be linked with liver injury and with biochemical alterations in jaundiced serum, but the pathogenetic mechanisms are still unclear. Cholestasis disarranges the processes of nonspecific and specific immunity and induces the tissue damage, with consequent liver regeneration, which is different from that seen after resection of the liver. Owing to our previous finding that peptidoglycan monomer linked with zinc (PGM-Zn) may improve some states of immunodepression and prolong the survival of cholestatic mice, in this study we attempted to investigate the hepatic changes after common bile duct ligation (CBDL) at the signal transduction level. For this purpose the priming, progression and inhibitory stage of liver regeneration were investigated by determination of protein products of protooncogenes (c-fos, c-jun, H-ras, K-ras, p53 and Ki67) 2, 4, 6, 8, 12 and 24 hours after CBDL, using the immunohistochemical methods in: 1) cholestatic mice pretreated with PGM-Zn before CBDL and in 2) cholestatic mice pretreated with PBS. The observed changes were also compared with the kinetics of compensatory liver growth (CLG) which follows after partial hepatectomy. The data have shown that: 1) the postcholestatic type of liver regeneration in several points differs from CLG (later expression of c-fos and earlier expression of H-ras and K-ras with maximum of p53 and Ki67 expression at 12 hours after BDL) and 2) that PGM-Zn induces earlier c-fos expression and pronounced K-ras expression in hepatocytes on 12 hours after BDL in comparison with PBS-treated group of mice. Peak of H-ras expression was moved to later terms, and some changes in tissue distribution of Ki67 were seen.
