MODELLING OF THE ALCOHOL DEHYDROGENASE PRODUCTION IN BAKER´S YEAST CULTIVATION
Baker's yeast (Saccharomyces cerevivisiae) is a very important source of different metabolites, especially enzymes, which are producing under different conditions1. The production of enzyme alcohol dehydrogenase by baker's yeast is directly connected to dissolved oxygen concentration and carbon source2, 3. Therefore optimising of these conditions and developing a model to describe the production of ADH was the aim of this work. Baker's yeast was cultivated in batch bioreactor at different initial concentration of glucose. The experiments were carried out under temperature of 30 oC and pH = 5. Dissolved oxygen concentration was set constant during the experiment at the value of 10-20 % concentration of saturation. The time dependent changes of biomass, glucose, ethanol, protein concentration, as well as ADH volume activities have been monitored. The unstructured model was chosen for describing kinetic of baker's yeast growth and consumption of substrate. The linear connection between baker's yeast growth and production of ADH was observed. Model parameters were estimated using package program SCIENTIST. Model validation has shown that proposed model describes the ADH production very well.
