Antiproliferative Effect of Polyphenols and Red Wines on Several Human Tumour Cell Lines

Polyphenols are a large group of compounds with a benzene core and attached hydroxyl groups. They can be found in plant origin foodstuffs and are known for their antioxidant properties. The goal of this research was to investigate the effect of gallic acid (GA), 4-methyl catechol (4-MC), L-3, 4-dihidroxyphenylalanine (L-DOPA), (± ; ; ; )- catechin (CAT), epicatechin (ECAT) and chlorogenic acid (CGA) as well as the effect of different concentrations of local red wines on the growth of tumour and normal cells in vitro. Citotoxicity of polyphenolic derivatives and wine concentrates was tested by MTT-assay and the effects of wine concentrates on proliferation of tumour cells were assessed by radioactive substrate incorporation in DNA, RNA and proteins. Phenolic content in wines used was assessed according to Singleton and Rossi method and ethanol content in the concentrates by MS-GC method. Results of the citotoxicity test showed that polyphenolic derivatives, GA and 4-MC were most effectively, inhibit cell growth in the concentration 10-4 M. Red wines have better antitumor effects than do individual substances. Colon carcinoma cells (CaCO-2, HT-29), as well as colon carcinoma metastasis (SW620) were the most effected by wine in concentrations 25% and 12.5% v/v. The concentrate with the greatest polyphenol content was shown to be the most effective. Biosynthesis assay of DNA, RNA and proteins indicates a standstill in cell growth, but also an increased protein synthesis. Our results indicate that polyphenols and polyphenol-rich foodstuffs have potential protheraphy effects on transformed cells.
