Lamotrigine concentrations in the serum and the brain tissue in rats

Purpose: The investigation of drug concentration in the serum and at the site of the action could be of importance in the understanding of possible mechanisms of drug action in different parts of the brain. The aim of this study was to investigate LTG concentrations in the rats' serum, hippocampus and cortex. Method: LTG concentrations were measured at different time-points (0 ; 0.5 ; 1 ; 2 ; 3 ; 6 h) in the rats' serum, hippocampus and cortex after intraperitoneal (i.p.) administration of 30 mg/kg. LTG concentrations were determined by high-performance liquid chromatography (HPLC) and quantifications were obtained by the external standard method using UV detection at 306 nm. Results: LTG concentrations were maximal 30 min after i.p. administration with the average value of 3.21± , .... 0.16 microg/ml in serum, 1.56± ; ; ; ; ; ; 0.12 microg/g in the cortex tissue and 2.00± ; ; ; ; ; ; 0.13 in the hippocampus. After that, LTG concentrations decreased continuously in the serum and the brain tissue, more rapidly in the serum (p<0.05). Drug concentrations in the cortex at all time-points were significantly lower than in the hippocampus (p<0.05). Conclusion: Our results show the correlation between LTG concentrations in the serum and the brain cortex and the hippocampal tissue. It is also obvious that LTG is accumulated in higher concentrations in the brain hippocampal tissue than in the cortex at all time points after the drug administration. This could be a contribution to the understanding of LTG mechanisms of action in epilepsy.
