Recombination and antirecombination in UV-irradiated Escherichia coli: the role of ruvB, ruvC, and recG genes

The products of ruv and recG genes are known to be engaged in the late phase of DNA recombination and recombinational repair, i.e. in the processing of Holliday junctions. In this work we present the genetic evidence for the involvement of these gene products in an opposite, antirecombinogenic, process. The experimental system used consists of UV-irradiated Escherichia coli cells and a thermoinducible mutant of lambda prophage. In earlier experiments we have found a phenomenon related to the inhibition of recombination which manifests itself in dying cells (i) as a progressive loss of prophage thermoinducibility, and (ii) as a progressive loss of prophage ability to recombine with the homologous infecting phage. The inhibitory process develops in fully repair proficient cells, and depends on the functional recA and recB genes. Since recA and recB gene products act in early phases of DNA recombination and recombinational repair, it was interesting to assess the possible role of gene products acting late in recombination in the development of post-UV loss of prophage recombinogenicity. Thus, we extend here the genetic analysis of this process by using E. coli ruvB, ruvC, and recG mutants. The results show that these mutants exhibit the loss of prophage recombinogenicity at much reduced rate compared to the rec+ ruv+ cells. Taken together, our previous and present results suggest that the processing of Holliday junctions during unsuccessful RecBC(D)-dependent recombinational repair of cell chromosome gives some recombination intermediates which, when reach the prophage, may abolish its ability to take part in further recombination reactions.
