The effect of liposomes with superoxide dismutase on the cells exposed to oxidative stress

Superoxide dismutase (SOD), antioxidative enzyme and potential antiinflammatory agent, was encapsupated into liposomes in order to increase its releasing time and to facilitate its cellular penetration. Positively, neutrally and negatively charged liposomes were prepared by the proliposome method using soybean lecithin, stearylamine and phosphatidylglycerol. The efficiency of SOD entrapment was 39-65%. A2128, human lung carcinoma cells, were treated with liposome-encapsulated SOD, 20 U/ml of media, for 24 hours. Positively, neutrally and negatively charged liposomes successfully delivered SOD to the cells. The biggest increase in the activity of cellular SOD was achieved by negatively charged liposomes (208% of the control). Neutral liposomes increased cellular SOD (183% of the control), but were not physically stable, and they agglomerated and settled on the cells. Positively charged liposomes increased cellular SOD (185% of the control), but were cytotoxic, as shown by the significantly increased lactate dehydrogenase leakage from the cells. The liposomes bearing negative charged successfully increased cellular SOD of CHO, Chinese hamster ovary epithelial cells (158% of the control) but not of THP-1, human monocyte leukaemia cells after 24 hours of treatment. Highly efficient SOD-loaded liposomes were produced using proliposome method. These liposomes prove to be successful in delivering SOD to the epithelial cells.
