SUPEROXIDE DISMUTASE IN LIPOSOMES: TREATMENT OF CELLS EXPOSED TO OXIDATIVE STRESS

Superoxide dismutase (SOD) was encapsulated into liposomes in order to increase its releasing time and to facilitate its cellular penetration. Anionic liposomes were prepared by the proliposome method using soybean lecithin and phosphatidylglycerol. The efficiency of SOD entrapment was 65%. Human lung carcinoma cells, A2182, were treated with liposome-encapsulated SOD, 20 U/ml of media for 24 hours. This treatment caused an increase in cellular SOD activity by 108%. Untreated, control cells and the cells treated with liposomal SOD were then exposed to oxidative stress caused by xanthine/xanthine oxidase for 4 or 24 hours. Oxidative stress did not affect cellular SOD activity, but caused an increase in cellular glutathione, a sensitive marker of cellular response to superoxide radical, in untreated cells. In the cells pre-treated with liposome-encapsulated SOD no increase in glutathione was detected. Treatment of A2182 cells with liposome-encapsulated SOD successfully increased cellular SOD and protected the cells from oxidative stress. Liposome-encapsulated SOD seems to be a promising alternative to conventional therapies in the treatment of oxidative stress-related diseases.
