The role of perforin (P) mediated cytotoxicity in the prevention of autoimmune destruction in the female reproductive tract
Molecular mechanisms of early cell-mediated cytotoxicity (CMC) are mediated by perforin (P) and Fas L. The role of P mediated pathway was investigated in experiments with deficient mice, and analyses of peripheral blood (PBL) and decidual (DL) lymphocytes in the human pregnancy. Mice simultaneously double deficient (Fas L and P) develop strong hysterosalpingitis and are infertile. However, mice homozygous for Fas L deficiency, but heterozygous at the P locus are fertile and protected from early death. This points to the potential role of P mediated pathway in the control of autoimmune destruction. Clinical investigations clearly showed that in the PBL in the first trimester normal human pregnancy an increase of CD4+ cells among P+ population was found, and significant decrease in pathological pregnancies. Contrary, drastic decrease in the PBL and almost complete absence in DL was found for CD16+P+ cells in the first trimester pregnancy, and significant upregulation at the end of the normal pregnancy and in the first trimester of pathological pregnancies. Decidual adherent cells (DAC), both stromal cells and macrophages, are indispensable for the maintenance of the P level in the DL. This mechanism is hormonally and cytokine (IL-15) regulated.
