Ochratoxin A induces changes of the amount of Hsp 70 in rat liver and kidney
We have examined the effects of ochratoxin A (OA) on the expression of heat-shock protein (Hsp 70), a member of highly conserved group of intracellular proteins whose synthesis is increased in response to variety of stress stimuli. Fisher rats were treated by two different doses of OA: 120 mikrograms/kg b.w./day for 10 days (group A, n=8) and for 35 days (group B, n=8), and 1200 mikrograms/kg b.w./day for 10 days (group C=8). The control group (group K) was represented by 8 untreated rats. Hsp 70 has been detected in liver and kidney homogenates by SDS-Page followed by immunoblotting using specific mAb agains Hsp 70 family. OA induced changes of the amount of Hsp 7o in both tissues, liver and kidney. In liver we detected the decrease of Hsp 70, that was dose/ and time/ dependent. In contrast to liver, in kidney it was found dose and time-dependent increase of Hsp 70. The role of heat-shock response to OA in prevention of the development of pathological changes related to OA exposure is subject to further studies.
